ATP7B codes for a copper-transporting ATPase — the pump that exports excess copper from liver cells into bile for elimination. When the pump works well, copper levels stay balanced; when it doesn’t, copper accumulates in the liver, brain, and other tissues. Severe loss of function produces Wilson’s disease; common variants produce a subtler version of the same imbalance.
Carriers of common ATP7B reduced-activity variants are associated with higher tissue copper accumulation and elevated copper-to-zinc ratios — which can show up as fatigue, brain fog, mood changes, or skin issues. The pattern often hides because serum copper can stay in range while tissue copper rises. Ceruloplasmin (the main copper-carrier protein) levels and copper-zinc ratios are more informative than serum copper alone.
Zinc directly competes with copper for absorption — adequate zinc intake suppresses copper uptake. Cooking in copper cookware adds dietary load. Drinking water from older plumbing can deliver substantial copper. Hormonal contraceptives, certain copper IUDs, and high-copper supplements all raise the load.
